Combinations of Recombinant Human Interferons and Retinoic Acid Synergistically

چکیده
The human acute promyelocytic leukemia cell line HL-6O is induced to differentiate into morphologically and functionally mature monocytelike cells by incubation with a combination of 10 nmol/L retinoic acid (RA) and various concentrations of recombinant immune interferon (rIFN-”y). These induced cells show marked increases in antibody-dependent cellular cytotoxicity (ADCC). antibody-coated erythrocyte (EA) rosettes, nonspecific esterase. and 5’-nucleotidase activity. rIFN-’y alone at concentrations of 1 0 to 1.000 U/mi has essentially no effect on morphological maturation. nitroblue tetrazolium reduction, and immunophagocytosis. However, rIFN-’y at these concentrations increases EA rosetting in a concentration-dependent manner that is not affected by 10 nmol/i RA. At a concentration of 1,000

The human acute promyelocytic leukemia cell line HL-60 is induced to differentiate into morphologically and functionally mature monocytelike cells by incubation with a combination of 10 nmol/L retinoic acid (RA) and various concentrations of recombinant immune interferon (rIFN-gamma). These induced cells show marked increases in antibody-dependent cellular cytotoxicity (ADCC), antibody-coated e...
متن کاملIn Vitro Interaction of Recombinant Tumor Necrosis Factora and A1l Trans-Retinoic Acid With Normal and Leukemic Hematopoietic Cells
Both human recombinant tumor necrosis factor alpha (TNFa) and all-trans-retinoic acid (RA) inhibit the in vitro clonal growth of human myeloid leukemic cells. We investigated the in vitro interaction of TNFa and RA with normal and a variety of leukemic myeloid cells. With the promyelocytic HL-60 cells. TNFa (>2.5 U/mI) in combination with RA synergistically inhibited clonal growth; TNFa at lowe...
متن کاملCombinations of retinoic acid with either sodium butyrate, dimethyl sulfoxide, or hexamethylene bisacetamide synergistically induce differentiation of the human myeloid leukemia cell line HL60.
All-trans-retinoic acid (RA), sodium n-butyrate (NaB), hexamethylene bisacetamide (HMBA), and dimethyl sulfoxide (DMSO) induce differentiation of the human acute myeloid leukemia cell line HL60. In the clinic, RA, NaB, or HMBA induce complete or partial remissions. However, the achievement and maintenance of effective plasma concentrations and toxicity have been problems. These difficulties led...
